Item Bases para el control genético de la tasa de secado del grano post madurez fisiológica en líneas parentales y sus híbridos derivados en maíz(2017) San Marco, Ignacio; Gambin, Brenda; Borrás, LucasLa humedad de grano a cosecha es de vital importancia para la producción de maíz, ya que afecta la calidad de la semilla y los costos de producción. Uno de los principales objetivos del mejoramiento es reducir el contenido de agua del grano a cosecha para disminuir el impacto económico del secado artificial. Además del momento en el que tiene lugar la madurez fisiológica, la tasa de secado después de la misma tiene un alto impacto en la humedad a cosecha. La tasa de secado es de difícil fenotipificación a campo, por lo que asociar el secado con otros caracteres de fácil determinación es de gran utilidad. Asimismo, determinar las bases genéticas (QTL) de estos caracteres es de gran interés para el mejoramiento. Finalmente, evaluar estos caracteres y sus bases genéticas no sólo en líneas sino también en híbridos derivados es relevante para su aprovechamiento en mejora genética. Los objetivos de la presente tesis fueron (i) caracterizar fenotípicamente una población de líneas e híbridos derivados para tasa de secado y diversos caracteres potencialmente asociados con la tasa de secado (tiempo a antesis, stay green, largo y número de chalas), (ii) establecer la correlación entre la tasa de secado en líneas e híbridos, y su relación con los otros caracteres fenotípicos medidos, y (iii) estudiar las bases genéticas (QTLs) de la tasa de secado tanto en líneas como en híbridos. Para cumplir estos objetivos se sembró una población biparental de líneas recombinantes (RlLs, n=129) y sus híbridos derivados de la cruza por un tester (n=129) en dos ambientes (fechas de siembra temprana y tardía) en Venado Tuerto. La tasa de secado se determinó ajustando un modelo lineal a mediciones periódicas de humedad post-madurez fisiológica en cada genotipo. Para la detección de QTLs se utilizó un modelo multi-atributo multi-ambiente para el conjunto de genotipos con regresión por mapeo de intervalos compuestos (CIM) en base a un mapa con más de 3000 SNPs. La variabilidad fenotípica en líneas e híbridos para tasa de secado fue menor que para el resto de los caracteres, lo que determinó una baja heredabilidad. La influencia del ambiente en la tasa de secado fue muy importante. Se encontraron correlaciones significativas entre la tasa de secado y caracteres como el tiempo a antesis y el stay green en la fecha de siembra tardía, lo que sugiere que ciertos atributos del genotipo serían importantes en ambientes que limiten la desecación. No se pudo establecer una relación entre las líneas y sus híbridos derivados para la tasa de secado. Se encontraron QTLs para algunos caracteres estudiados como tiempo a antesis, y número de chalas, y sólo uno para tasa de secado en líneas, aunque no fue consistente entre ambientes. Se concluye que la mejora genética para tasa de secado se ve limitada por su difícil fenotipificación y fuerte interacción con el ambiente. La elección de la población de mapeo es asimismo sumamente importante para asegurar el éxito de los resultados en este tipo de atributos.Item Detección de QTLs para mecanismos fisiológicos relacionados con la determinación del número de granos de sorgo(2014) Spagnolli, Florencia Carla; Borrás, Lucas; Gambín, BrendaEl número de granos (NG) constituye el componente que mejor explica la variabilidad en rendimiento para el cultivo de sorgo. El objetivo de este proyecto fue estudiar las bases genéticas (QTL) de este caracter en una población de 250 RILs F5 a partir de atributos secundarios ligados con su determinación. El modelo ecofisiológico utilizado considera que el NG depende de la tasa de crecimiento del cultivo alrededor de floración (TCCa), de la proporción de biomasa particionada a los órganos reproductivos (PR) y de la cantidad de granos fijados por unidad de crecimiento reproductivo (EfG). Estos atributos se evaluaron durante dos condiciones ambientales. Existió variabilidad fenotípica para todos los caracteres estudiados (p<0,001). Variaciones en el NG estuvieron positivamente asociadas a variaciones en PR y EfG. Estos dos caracteres presentaron una alta heredabilidad en sentido amplio. Se detectaron un total de 12 QTL (LOD≥2,5), algunos de ellos consistentes entre ambientes para PR y EfG. Los QTL detectados explicaron entre 15 y 40% de la variabilidad observada. Este es el primer estudio sobre las bases genéticas en partición de biomasa y determinación de NG en sorgo, y constituye un paso relevante para entender la determinación de los factores genéticos detrás del NG y rendimiento del cultivo.Item Detección y caracterización de QTLs para mecanismos fisiológicos relacionados con el peso final de los granos, sus patrones de crecimiento y con el secado de los granos de maíz(FCA-UNR, 2018) Cresta, Agustín Eduardo; Borrás, Lucas; Pizarro, GuillermoLos cambios operados en los últimos años en la estrategia de siembra del cultivo de maíz en diferentes regiones ubica el período entre floración y madurez comercial en condiciones climáticas contraproducentes para la pérdida de humedad de los granos. El período de madurez de loa granos de maíz, se dividió en dos etapas, primero desde floración hasta madurez fisiológica, donde se define el peso de grano (PG), y segundo la etapa de secado de granos, donde se define la humedad del grano (HG) a cosecha. El objetivo de la presente tesis fue estudiar las bases genéticas (QTL) de la maduración de los granos de maíz en una población de 173 líneas doble haploide (DH). Los caracteres evaluados se descompusieron en atributos simples, y existió variabilidad fenotípica para todos los caracteres estudiados (p<0,001). También se observó que genotipos con tiempo térmico (TTA) similares, pueden tener duraciones de llenado de grano (DLLG) muy diferentes. No existe una correlación significativa entre el contenido de humedad de los granos a madurez fisiológica (CHMF) y la duración del secado los granos (DSG), por lo tanto, los genotipos pueden tener diferentes DLLG y valor similares DSG. Se observó que a medida que el CHMF es mayor, la Tasa de secado de granos (TSG) también lo es. Esto podría indicar, que el CHMF condiciona en parte a la TSG. Se detectaron 29 QTL (LOD≥2,5). La variación fenotípica explicada (VFE) por carácter, vario desde 7 a 60%. La ubicación conjunta de QTL para PG, DLLG, tasa de crecimiento de los granos (TCG), máximos contenido de agua en los granos (MCAG), CHMF, TSG y DSG, ayudan a explicar las correlaciones fenotípicas observadas. Los resultados obtenidos aportan al conocimiento actual de las bases genéticas de la maduración los granos de maíz a través del análisis fenotípico y la identificación de QTL de caracteres asociados a la determinación del PG y el secado de los granos.Item Pérdida de humedad a campo bajo condiciones de secado contrastantes en diferentes genotipos de maíz(2017) Mendive, María Eugenia; Borrás, LucasEn fechas de siembra tardías de maíz el secado del grano es un atributo muy relevante al momento de elección del genotipo a sembrar. La pérdida de humedad del grano es muy lenta debido a que la humedad relativa media del aire es alta, y dificulta el secado a campo de la espiga. La tasa de pérdida de humedad y la madurez relativa del genotipo son dos características de suma importancia en la determinación del contenido de humedad a cosecha. El objetivo del presente trabajo fue estudiar el efecto del genotipo, del ambiente y la interacción genotipo x ambiente para pérdida de humedad del grano en fechas de siembra contrastantes (septiembre y octubre vs. diciembre). Los atributos rendimiento, contenido de humedad del grano a comienzo del secado, tasa de secado diaria y humedad a cosecha, fueron medidos en siete híbridos comerciales, en dos localidades, durante dos años consecutivos, para fechas de siembra temprana y tardía. Existió variabilidad fenotípica para todos los caracteres estudiados. La fecha de siembra fue el factor que más afectó la tasa de secado del grano y el rendimiento logrado, y explicó el 60 y el 33 de la variación total observada en ambos caracteres, respectivamente. No se observaron cambios de importancia en el ranking de híbridos para tasa de secado, es decir, los híbridos de mayor velocidad de secado en siembras tempranas fueron los de mayor tasa de secado en siembras tardías. Este es un resultado de alto impacto para los programas de mejoramiento que estudian el contenido de humedad del grano a cosecha.
